Born in Newcastle upon Tyne, England in 1883, Wilson came to Australia as a child the following year. After schooling, she studied at the National Gallery art school in 1901-06 under Bernard Hall and Frederick McCubbin. She first exhibited in 1913-14 in an exhibition of 'The Twelve Melbourne Painters' with a group of her fellow ex-students. In 1927 Wilson travelled overseas for two and a half years visiting Europe and Britain and in 1928 she exhibited at the Beaux Arts Gallery, London, and next year at Australia House. In the 1930s Dora Wilson painted a series of 19th century Australian historical pictures, and these were exhibited in Melbourne in 1935. A foundation member of the Australian Academy of Art, she was known as a painter of street scenes and pastel nude studies.
